Atomic force microscopy (AFM) allows dynamic detection of the adhesion or affinity between the sample surface and the cantilever. This feature can be used to select aptamers that bind to their targets with very high affinity. By combining SELEX and AFM methods, aptamers with strong affinity for the target molecule can be obtained.

AFM can work in vacuum, ultra-high vacuum, gas, solution, electrochemical environment, normal temperature and low temperature, so it has a wide range of applications. In biology, atomic force microscopy can be used to study the structure and other properties of biological macromolecules, such as the observation of proteins, RNA, DNA, and even cells and viruses.
In the selection method using AFM, aptamers with high affinity for the target molecule can be obtained in only three replicate rounds. Thus, the AFM SELEX strategy is a very useful method for obtaining aptamers quickly. In this new SELEX strategy using AFM, only the oligonucleotide bound to the target is retained on the chip, while the non-specific binding is reduced on the chip surface, resulting in rapid acquisition of aptamers with high affinity for the target.
