Although nucleic acid aptamers targeting purified proteins have a wide range of applications, the screening process is complicated and generally requires more than ten rounds of screening, which consumes a lot of manpower, financial and time resources, and the resulting nucleic acid aptamers may also have low affinity and poor specificity, which affect their subsequent applications.

Due to the structural and chemical properties of aptamers, there are theoretically a wide variety of targets, and aptamer screening techniques have been developed for non-purified proteins as targets, among which the more representative ones are cell SELEX and in vivo SELEX.
