Elevated expression levels of cell surface proteins are often a marker of disease response and the basis for the development of specific molecular probes. Cell surface proteins are primarily involved in signal transduction and ligand binding. Cell surface receptor molecules generally respond to altered biological activity in disease states by exhibiting different levels of expression. It is this differential expression that allows researchers and clinicians to distinguish diseased cells from normal cells.
Nucleic acid aptamers can be well suited for the function of recognition of cell surface markers. Successful selection of high-affinity aptamers depends on whether the targeting epitope is disturbed. Therefore, it is necessary to maintain the correct structure of the desired protein during SELEX in order to successfully obtain high-affinity aptamers. Researchers developed a variant of the original SELEX, called complex target SELEX.
The targets of classical SELEX screening are mostly single proteins or small molecules, and the oligonucleotide sequences screened by these targets are limited to a few families, and the ligands of these families often compete with each other for the same locus or crossover sites of the targets. In contrast, complex target SELEX technology targets not pure proteins or small molecules, but a mixture of substances, which greatly increases the applicability.

It is actually using the same oligonucleotide library for multiple screening at the same time, the specific ligands of different targets in the mixture will be screened out at the same time without interfering with each other.
Complex target SELEX can isolate aptamers targeting a wide range of cell surface proteins, including receptors and markers of cell differentiation, and determinants of disease in pathogenic organisms, and thus plays an important role in a wide range of therapeutic and diagnostic applications.
In 1998, researchers first applied SELEX technology to the screening of total human erythrocyte proteins by using the same oligonucleotide library to simultaneously screen for specific aptamers of different targets in a mixture without interfering with each other. In a subsequent study, researchers used recombinant proteins overexpressed on the surface of mammalian cells as targets for screening and successfully obtained aptamers with Kd values down to the nmol/L level.
