The demands for separation of chiral compounds are continuously increased in fine chemical/biological and pharmaceutical industry. One of the main concerns in chiral separation is that each enantiomer of a racemic drug may have different pharmacokinetic and pharmacodynamic effects due to their differential metabolism.
Nucleic acid aptamers screened with R- or S-type molecules can bind chiral molecules of that type and distinguish their enantiomers with different spatial structures. Nucleic acid aptamers of chiral molecules will have great potential for chiral separation and detection of natural polysaccharides and derivatives, macrocyclic antibiotics, chiral macrocyclic ligands, ligand exchange complexes, and chiral surfactants in the biological and chemical pharmaceutical industries.
Researchers have screened five DNA sequences using isobutylpropionic acid racemate ibuprofen mixed in a 1:1 substance ratio as the target. Three of the DNA sequences showed affinity for both racemate and R-isobutylpropionic acid. The other 2 DNA sequences had affinity for both racemate and S-isobutylpropionic acid. The above five nucleic acid aptamers can be used for the in vivo detection of ibuprofen in pharmacokinetic analysis and the isolation of ibuprofen chiral drugs.
Researchers have immobilized L-tryptophan on agarose microbeads and selected nucleic acid library was firstly reverse screened with L-tyrosine as the target and then interacted with L-tryptophan. DNA nucleic acid aptamers specifically recognizing L-tryptophan were obtained after 12 rounds of screening and could be used for chiral disassembly of D/L-tryptophan.
